[体外研究]

L-组氨酸完全抑制生长,其对生存力的影响与FLO11表达成反比。 L-组氨酸不影响Δflo11和S288c菌株的活力。 L-组氨酸显着降低了flor酵母的气液生物膜形成和与聚苯乙烯的粘附,而对FLO11基因的转录水平没有影响。此外,L-组氨酸可以改变弗洛尔酵母细胞壁上的几丁质和聚糖含量[1]。

[体内研究]

L-组氨酸(100 mg/kg)完全抑制硫代乙酰胺治疗大鼠的脑水肿[2]。在L-组氨酸饮食组中由来自下丘脑的高K +刺激的组胺释放是对照组的60%。然而,L-组氨酸饮食不会改变其他单胺及其代谢物的浓度。野外试验显示,L-组氨酸饮食组在中心区域花费的时间较短,而明/暗盒测试表明L-组氨酸饮食组在灯箱中花费的时间较短,提示L-组氨酸饮食可引起焦虑样行为[3]。

[动物实验]

大鼠:每天给动物施用TAA(300mg / kg ip)3天。在每次TAA注射前2小时,将L-组氨酸(100mg / kg)溶解在盐水中并每天注射(ip)。为防止低血糖和脱水,每12小时给予大鼠12.5ml / kg的液体治疗(5%右旋糖和0.45%盐水,20mEq / L氯化钾),sc标准对照接受盐水(用于TAA的载体),而另一组大鼠每天单独给予L-组氨酸(100mg / kg),持续3天。对TAA治疗的大鼠进行临床监测,并对脑病的阶段进行分级[2]。小鼠:对照组喂食含有5.08克L-组氨酸/ kg的AIN-93G纯化饮食,而L-组氨酸饮食组喂食含有1.28克L-组氨酸/ kg的AIN-93G(25% AIN-93G中的组氨酸含量。为了平衡氨基酸的总量,将谷氨酰胺添加到L-组氨酸饮食中以抵消组氨酸含量的变化(18.32g L-谷氨酰胺/ kg AIN-93G对比23.72g L-谷氨酰胺/ kg L-组氨酸饮食)。两种饮食都是等氮的。在8周龄时,将小鼠称重并分配给2种不同的饮食。允许小鼠随意获取水及其各自的饮食,并且在开始实验之前将它们在实验室中饲养至少2周[3]。

1.从猪血、牛血中提取。猪血经喷雾干燥制得血粉,每100kg猪血约18kg血粉。L-组氨酸常用其盐酸盐([7048-02-4])。将含有L-组氨酸的洗脱液,浓缩至出现结晶,趁热用盐酸调至pH为2.5,立即加入2倍于溶液量的乙醇,静置,沉淀,过滤,即得L-组氨酸盐酸盐粗品,经脱色、重结晶、干燥得成品。L-组氨酸也可从脱脂大豆的水解产物中提取。
