[体外研究]

在基于细胞的报告基因检测中,GW 6471完全抑制GW409544诱导的PPARα活化,IC50为0.24μM[1]。通过MTT测定评估PPARα在肾细胞癌(RCC)细胞活力中的功能作用。将Caki-1(VHL野生型)和786-O(VHL突变)细胞分别与特异性PPARα激动剂WY14,643或特异性PPARα拮抗剂GW6471一起孵育,浓度为12.5至100μM,持续72小时,评估细胞活力。虽然WY14,643对细胞活力没有影响或略有增加,但GW 6471在两种细胞系中显着且剂量依赖性地抑制细胞活力(高达约80％)[2]。

[体内研究]

为了测试体内PPARα拮抗作用的抗肿瘤活性,使用皮下异种移植小鼠模型。将Caki-1细胞皮下植入裸(Nu/Nu)小鼠中。在肿瘤块直径达到约5mm后,每隔一天腹膜内注射GW6471,持续4周,剂量(20mg/kg小鼠体重),这被描述为在体内剂量反应研究中有效并在此确认要有效。在载体和GW6471处理的动物之间肿瘤生长存在显着差异。基于动物的重量,在GW6471的剂量下未观察到毒性,并且实验室值(包括肾和肝功能测试)未受到不利影响。为了证明GW 6471的靶向作用,在肿瘤中评估了c-Myc水平,其显示GW 6471处理的动物显着减少[3]。

[细胞实验]

将786-O和Caki-1细胞接种在96孔板中。将两种细胞与WY14,643或GW6471分别以12.5至100μM的浓度孵育72小时，并且在指定的处理后，将细胞在MTT溶液/培养基混合物中孵育。然后，除去MTT溶液，将每个孔中的蓝色结晶沉淀物溶解在DMSO中。使用酶标仪[2]量化每个孔在540nm处的可见吸光度。

[动物实验]

小鼠[3]雄性无胸腺Nu / Nu小鼠（8周龄，~25g体重）在侧腹区域皮下注射1×105个Caki-1细胞（3：1DMEM-基质胶）。通过卡尺每周监测肿瘤进展。当肿瘤大小达到~80-100mm 3时，将动物随机分成四组并开始治疗（第1天）。载体组通过口服强饲法接受腹膜内DMSO（PBS中4％）和植物油。 PPARα组每隔一天腹膜内注射相同载体中的GW6471（20mg / kg体重;其他地方报道鼠剂量反应）。舒尼替尼组通过口服强饲法（40 mg / kg体重）在植物油中接受舒尼替尼5天/周。另一组接受GW 6471 + Sunitinib。为了确定治疗的任何潜在毒性，测量动物的体重并监测不良反应的迹象。在第28天，使小鼠安乐死并测定肿瘤质量。计算肿瘤生长率[3]。
