[体外研究]

咖啡酸对组胺诱导的反应具有抑制作用,并且当用于预处理的浓度从0.1增加至1mM时,咖啡酸的抑制作用逐渐增加,类似于典型的剂量依赖性反应。用1mM咖啡酸预处理HEK293T-TRPV1细胞导致辣椒素诱导的反应的显着抑制。当使用较低浓度的咖啡酸时,辣椒素诱导的反应的抑制作用不太明显。钙成像实验表明,咖啡酸孵育导致组胺敏感的背根神经节(DRG)神经元的显着抑制。用咖啡酸(1mM)预处理导致响应性DRG神经元对组胺应用的百分比从12.5%显着降低至2.1%。用1mM咖啡酸预处理显着阻断了表达TRPA1的细胞中的烯丙基异硫氰酸酯(AITC)诱导的细胞内钙增加。咖啡酸还能够阻断AITC诱导的TRPA1活化[1]。

[体内研究]

用咖啡酸(500mg/kg)预处理的小鼠表现出显着较少的组胺诱导的刮擦(30.50±10.87次/ 1小时,n = 6)。进一步发现,较低剂量的咖啡酸(100 mg/kg)在组胺诱导的刮擦中的抗刮擦效果方面没有显着效果,尽管似乎有减少的趋势(49.40±12.35次/ 1) h,n = 5)。用500 mg/kg咖啡酸预处理可显着抑制氯喹诱导的搔抓(161.6±31.42次/ 1 h,n = 5)[1]。咖啡酸显着降低5-LO mRNA的表达(P <0.01)剂量依赖性地在海马中。与缺血再灌注(I/R)未治疗组相比,I/R-咖啡酸组中5-LO蛋白表达显着降低(P <0.05或P <0.01),尤其是I/R-咖啡酸组(50 mg/kg)。与I/R未治疗组相比,低剂量和高剂量咖啡酸组的寻找平台潜伏期显着缩短,I/R-咖啡酸组缩短平台潜伏期最明显(50 mg/kg)(P <0.01)。在低剂量咖啡酸组中,细胞损伤仍然明显,核固缩率为(63.6±2.8)%,而在高剂量咖啡酸组中,海马神经元核固缩显着减少,核固缩率为(13.3±) 3.0)%[2]。

[细胞实验]

为了确定细胞活力,进行MTT测定。 HEK293T细胞在37℃,一天前在96孔板中培养,使得实验当天细胞的汇合为85%至90%。在实验当天,用不同浓度的咖啡酸处理细胞10分钟。对照细胞仅用培养基处理。除去上清液并用PBS洗涤后,将MTT试剂(5mg / mL)直接加入到新鲜培养基中。然后将细胞在37℃下再孵育4小时,然后排出培养基并在黑暗条件下过夜储存。第二天,将DMSO加入每个孔中并在振荡器中混合10分钟,之后使用微板记录仪在490nm处读取板,参比波长为620nm。相对细胞存活率(%)表示为相对于未处理对照细胞的百分比[1]。

[动物实验]

将大鼠分为五组:假手术组(n = 9),缺血再灌注(I / R)未治疗组(n = 9),I / R-咖啡酸组(10mg / kg)(n = 9),I / R-咖啡酸组(30mg / kg)(n = 9)和I / R-咖啡酸组(50mg / kg)(n = 9)。在I / R-咖啡酸组中,在缺血前30分钟通过腹膜内注射给予大鼠10,30,50mg / kg(用0.3%羧甲基纤维素钠制备)的咖啡酸。假手术组和I / R组用等体积的0.3%羧甲基纤维素钠治疗[2]。
